- PROBLEM TO BE SOLVED: To provide a simpler and efficient method for judging carrot cytoplasmic type based on the difference of mitochondoria DNA structure. SOLUTION: This method for assessing carrot cytoplasmic type and a kit to be used therefor comprise the following steps; (1) an oligonucleotide substantially identical with or substantially complementary to part of a DNA specific to normal-type cytoplasm, DNA specific to petaloid-type male-sterile cytoplasm, or DNA specific to Brown answer-type male-sterile cytoplasm, is synthesized; (2) a solution containing the above oligonucleotide, the mitochondoria DNA of a carrot intended for assessing its cytoplasmic type, DNA polymerase, and dNTP, is heated several times; and (3) the solution is subjected to electrophoresis, and based on the resulting electrophoretic pattern, the aimed cytoplasmic pattern is judged. This method enables carrot cytoplasmic type to be easily and efficiently judged, and has such effect as to enable the parent root of F1 variety to be selected early.
